Lorus Therapeutics Announces Publication on Drug Levels and Clearance Rates for Anticancer Drug LOR-2040

- Investigation by a Clinical Research Team at Ohio State University - -

TORONTO, CANADA - September 9, 2008 - Lorus Therapeutics Inc, (TSX: LOR, AMEX: LRP) (“Lorus”), a biopharmaceutical company specializing in the research and development of pharmaceutical products and technologies for the management of cancer, today announced a scientific publication by a clinical research team at The Ohio State University (OSU) College of Pharmacy entitled “Enzyme Kinetics of GTI-2040, a Phosphorothioate Oligonucleotide Targeting Ribonucleotide Reductase” (Drug Metabolism and Disposition 2008, July 24 Epub ahead of print).

The primary author, Dr. Xiaohui Wei, presents results on the metabolism and clearance of GTI-2040, now known as LOR-2040. These findings are reported from a clinical research team involved in the clinical trial of LOR-2040 and high dose cytarabine for treatment of refractory and relapsed acute myeloid leukemia under the overall direction of Dr. Guido Marcucci as Principal Investigator and Dr. Kenneth Chan, Co-Director of the PharmacoAnalytic Core Laboratory at the OSU Comprehensive Cancer Center.

This investigation identified complex factors including activity of liver microsomes that together predicted the circulating steady state drug levels and clearance rates observed for LOR-2040 in the clinical trial. Previously reported data with LOR-2040 demonstrated significant uptake of drug in bone marrow blasts and downregulation of the R2 cellular target of LOR-2040 that correlated with clinical response.

These investigations at OSU were supported by a National Institutes of Health (NIH) grant and National Cancer Institute (NCI), Cancer Therapy Evaluation Program (CTEP) sponsorship of the clinical program. Lorus is collaborating in this program with the Division of Cancer Treatment and Diagnosis, NCI on the development of LOR-2040.

LOR-2040 (formerly GTI-2040) is an RNA-targeted drug that specifically targets the R2 component of ribonucleotide reductase, which is required for DNA synthesis and cell proliferation. Through downregulation of R2, LOR-2040 has demonstrated strong antitumor and antimetastatic activity in a variety of tumor types in both in vivo and in vitro models and is under study in a multiple Phase I/II clinical program, including an advanced Phase II clinical trial with LOR-2040 and high dose Ara-C (HiDAC) in refractory and relapsed Acute Myeloid Leukemia (AML). The R2 target has been described as a malignant determinant that is elevated in a wide range of tumors, which can cooperate with a variety of cellular cancer causing genes known as oncogenes to enhance tumor growth and metastatic potential.
